Strain typing and determination of population structure of Candida krusei by multilocus sequence typing.

چکیده
A multilocus sequence typing (MLST) scheme for Candida krusei was devised, based on sequencing of six gene fragments of the species. The existence of heterozygous results for each of the six fragments sequenced confirms that C. krusei is diploid for at least part of its genome. The C. krusei MLST scheme had a discriminatory index of 0.998, making this system ideal for strain typing of C. krusei clinical isolates. MLST data for 122 independent C. krusei isolates from a range of geographical sources were analyzed by eBURST, structure, and the unweighted-pair group method using average linkages to derive a population structure comprising four subtype strain clusters. There was no evidence of geographical associations with particular subtypes. Data for pairs of isolates from seven patients showed that each patient was colonized and/or infected with strain types that were indistinguishable by MLST. The C. krusei MLST database can be accessed online at http://pubmlst.org/ckrusei/.

Rapid development of Candida krusei echinocandin resistance during caspofungin therapy.
In invasive candidiasis, there has been an epidemiological shift from Candida albicans to non-albicans species infections, including infections with C. glabrata, C. parapsilosis, C. tropicalis, and C. krusei. Although the prevalence of C. krusei remains low among yeast infections, its intrinsic resistance to fluconazole raises epidemiological and therapeutic concerns. Echinocandins have in vitr...
متن کاملMultilocus Sequence Typing of the Clinical Isolates of Salmonella Enterica Serovar Typhimurium in Tehran Hospitals
Background: Salmonella enterica serovar Typhimurium is one of the most important serovars of Salmonella enterica and is associated with human salmonellosis worldwide. Many epidemiological studies have focused on the characteristics of Salmonella Typhimurium in many countries as well as in Asia. This study was conducted to investigate the genetic characteristics of Salmonella Typhimurium using m...
متن کاملMolecular-Genetic Approaches for Identification and Typing of Pathogenic Candida Yeasts: A Review
Candida is a medically important fungi because of its high frequency as a commensal and pathogenic microorganism causing superficial as well as invasive infections. Because the accurate diagnosis of candidiasis remains difficult, a fast and reliable assay for characterization of fungal pathogens is critical for the early initiation of adequate antifungal therapy and/or for introduction of preve...
متن کاملMultilocus Sequence Typing (MLST) of Enterococcus faecalis clinical isolates in Ilam, Iran
Introduction: The Enterococcus faecalis (E. faecalis) is the one of the pathogenic bacteria that become famous and considerable in the recent years. Here we tried to do typing the E. faecalis isolates to provide advantageous information that can help us to understand epidemiological communication between the E. faecalis isolates. Materials and methods: One hundred E. faecalis were isolated fr...
متن کاملGenetic Heterogeneity among Leishmania major Isolates in Iran Determined by Restriction Fragment Length Polymorphism (RFLP) and Multilocus Microsatellite Typing (MLMT)
Background & Aims: In recent years, molecular methods for characterizing genetic heterogeneity have found a major place in modern approaches. In this study, two different molecular techniques including Restriction Fragment Length Polymorphism (RFLP) and Multi Locus microsatellite typing (MLMT) were carried out in order to evaluate genetic heterogeneity among isolates of Leishmania major in Iran...
